"Pharmacological effect Pentalgin extra-gel

Ketoprofen is a nonsteroidal anti-inflammatory agent intended for topical use. It effectively inhibits cyclooxygenase and also reduces the activity of lipoxygenase and bradykinin. Ketoprofen also stabilizes lysosomal membranes and prevents the release of enzymes involved in the inflammatory process. The main properties of the drug are pronounced analgesic, anti-inflammatory and antipyretic action. Ketoprofen does not impair articular cartilage.

Pharmacokinetics Pentalgin extra-gel

Ketoprofen in the form of a topical gel effectively penetrates the skin depending on the physical and chemical properties of its components. The rate of release and absorption of the gel depends on its composition. The relative bioavailability of ketoprofen when topical gel is used is about 5%, which is significantly less than with oral administration. Ketoprofen in the form of a gel has an effect mainly at the local level and does not have a systemic effect.

Distribution

Ketoprofen binds well to plasma proteins (99%) and penetrates joint tissues, including synovial fluid, reaching therapeutic concentrations. The concentration of ketoprofen in the blood is insignificant. After topical gel application, the maximum plasma concentration of ketoprofen is reached in 6 hours. Considerable concentrations are present in subcutaneous fat, periarticular tissues, and synovial fluid. The drug lasts in tissues for 12 hours.

Metabolism and elimination
Ketoprofen is metabolized in the liver to form conjugates, which are excreted primarily by the kidneys. The metabolic process of ketoprofen is independent of patient age, presence of severe renal insufficiency, or cirrhosis. Ketoprofen is gradually excreted by the kidneys.
